From your post, I can't determine if you have fuel leaking in the carb, and possibly hydrolocked the cylinder with gas, or the valves are out of adjustment, and can turn past compression stroke.
Look into the carb, and see if there is a pool of gas. If yes, then you need to remove the spark plug, and either let the gas evaporate, or blow out with compressed air ( do not crank the engine, because fuel will spray out, and can cause static electricity to ignite the fuel), remove and clean the carb, best down with a carb kit, check the oil for gas smell, and change if gas in oil.
If no fuel present, then remove valve cover, remove spark plug, turn flywheel to where piston is just past TDC, and both valves are closed. Adjust valves to .003-.005 intake and .005-.007 exhaust, and see if that fixes the problem.
