I have a new cub cadet pro Z148S with the dual ZT3400 transmissions. They have the internal expansion tanks. The manual says that they are maintenance free. My understanding was that the first service was to be done at 50 hours and then at every 250 hours thereafter. Any advice would be much appreciated.

Kinda contradictory info given. If the units has external filters follow the initial 75 hr then 400 hr intervals afterwards oil and filter change schedule. Internal filtered units are considered sealed with no maintenance schedule.
I have had both a Cub Cadet LTX1045 tractor with a sealed Hydro Gear transmission and a Cub Cade XT1 tractor with a sealed Tuff Torq transmission. On both I was able to remove the top fill (not vent) plug and use an evacuation pump to “suck” out about ¾ of the fluid inside. I then used a funnel to replace the fluid. You might want to consider that approach on sealed units.
